autologous anti-TM4SF1 CAR-T cells (also known as KQ-L6 or CART-TM4SF1) is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting Transmembrane 4 L6 family member 1 (TM4SF1). TM4SF1 is a cell surface antigen highly expressed in various solid tumors and tumor endothelial cells, playing a key role in tumor cell proliferation, migration, and angiogenesis. Developed by Shanghai Keqi Pharmaceutical Technology (Shanghai Ultra-T Immune Therapeutics), Novatim, and Shanghai 10th People's Hospital, this cell therapy is designed to recognize and eliminate TM4SF1-expressing tumor cells. The CAR construct typically incorporates a safety switch, such as a truncated human epidermal growth factor receptor (EGFRt), to allow for selective depletion of the CAR-T cells in the event of severe toxicity. It is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors, particularly digestive system cancers including colorectal, pancreatic, gastric, and liver cancers.
