Overview

Autologous B7-H3 CAR T is an investigational chimeric antigen receptor (CAR) T-cell therapy developed by the National Cancer Institute (NCI). It consists of a patient's own T cells that have been genetically engineered to express a CAR targeting B7-H3 (also known as CD276), a transmembrane protein that is highly expressed in various solid tumors but has limited expression in normal tissues. The therapy is designed to recognize and eliminate B7-H3-positive cancer cells. It is currently being evaluated in Phase I clinical trials for adult patients with extensive-stage small cell lung cancer (SCLC) and extrapulmonary neuroendocrine carcinoma (EPNEC), as well as pediatric patients with relapsed or refractory embryonal tumors, including neuroblastoma, rhabdomyosarcoma, Ewing sarcoma, and Wilms tumor. Treatment typically involves lymphodepleting chemotherapy with cyclophosphamide and fludarabine prior to the infusion of the modified T cells.
