Overview

This investigational cell therapy consists of autologous mesenchymal stem cells (MSCs) derived from the patient's own bone marrow, developed and studied by Antonio Uccelli and the MESEMS (MEsenchymal StEm cells for Multiple Sclerosis) trial network. The therapy involves the ex vivo expansion of these cells followed by a single intravenous infusion at a dose of 1-2 × 10^6 cells per kilogram of body weight. It is designed to treat multiple sclerosis by leveraging the innate immunomodulatory and neuroprotective properties of MSCs, which are thought to migrate to sites of inflammation and secrete paracrine factors that suppress pathogenic immune responses and support neural repair. The international Phase 2 MESEMS trial evaluated this therapy across several MS subtypes; while it demonstrated a favorable safety profile, it did not meet the primary efficacy endpoint of reducing gadolinium-enhancing lesions on MRI over 24 weeks.
