Autologous C7R.CD30-CAR-EBVSTs is an investigational cell therapy consisting of autologous Epstein-Barr virus-specific T lymphocytes (EBVSTs) genetically modified to express a chimeric antigen receptor (CAR) targeting CD30, a constitutively active interleukin-7 receptor (C7R), and an inducible caspase-9 (iC9) safety switch. The CD30-CAR enables the T cells to recognize and eliminate CD30-positive lymphoma cells. The inclusion of C7R provides a continuous survival and proliferation signal, which is intended to enhance the persistence and expansion of the T cells within the immunosuppressive tumor microenvironment. The iC9 safety switch allows for the rapid elimination of the infused cells in the event of severe toxicity. The use of EBVSTs as the cellular backbone is designed to leverage their inherent viral specificity for improved safety and longevity. This therapy is currently being evaluated in Phase I clinical trials (ANCILE-30) for patients with relapsed or refractory CD30-positive lymphomas, including Hodgkin lymphoma and various non-Hodgkin lymphomas.
