**Autologous cancer stem cell vaccine** is a personalized dendritic cell-based cancer vaccine generated from a patient's own tumor and immune cells. In the reported glioblastoma program, cancer stem cells are isolated from the patient's tumor biopsy, expanded in vitro as tumorspheres, and used as a source of amplified tumor stem cell mRNA, which is then transfected into autologous monocyte-derived dendritic cells; the final product is administered by intradermal injection to induce T-cell-mediated immunity against patient-specific cancer stem cell antigens thought to drive tumor propagation and recurrence. The approach has been studied primarily in glioblastoma, where early clinical experience suggested acceptable safety, induction of immune responses in vaccinated patients, and potential prolongation of progression-free survival, but it remains an individualized investigational immunotherapy rather than a marketed pharmaceutical product.
