Drug intelligence / Profile preview
autologous CAR-BCMA CAR-T cells is an investigator-initiated, autologous, point-of-care (POC) chimeric antigen receptor (CAR) T-cell therapy developed by Sheba Medical Center (led by Dr. Hila Magen) for the treatment of relapsed or refractory multiple myeloma. The CAR construct consists of a murine anti-BCMA single-chain variable fragment (scFv) derived from clone 11D5-3, a CD28 costimulatory domain, and a CD3-zeta signaling domain. Manufactured on-site at Sheba Medical Center using fresh, non-cryopreserved T-cells, this point-of-care approach significantly shortens the vein-to-vein turnaround time to approximately 10 to 11 days, reducing the need for bridging therapy. It is currently being evaluated in a Phase 1/2 clinical trial (NCT05243212) in patients with multiple myeloma who have progressed after prior lines of therapy.
