Overview

Autologous CCR4 CAR T cells are a cell-based immunotherapy consisting of a patient's own T cells genetically engineered to express a second-generation chimeric antigen receptor (CAR) targeting C-C chemokine receptor type 4 (CCR4). The CAR construct typically utilizes a lentiviral vector and incorporates a 4-1BB (CD137) costimulatory domain and a CD3-zeta signaling domain. CCR4 is a protein frequently overexpressed in various mature T-cell malignancies, including peripheral T-cell lymphoma (PTCL), cutaneous T-cell lymphoma (CTCL), and adult T-cell leukemia/lymphoma (ATL). By targeting CCR4, these CAR T cells specifically recognize and eliminate malignant cells. The therapy is currently being investigated in Phase I clinical trials for relapsed or refractory T-cell non-Hodgkin lymphomas.
