Autologous CD19-BCMA dual-target CAR T-cell therapy is a personalized cell-based immunotherapy developed by Beijing Boren Hospital for the treatment of relapsed or refractory autoimmune diseases. The therapy involves the genetic modification of a patient's own T cells to express chimeric antigen receptors (CARs) that target both CD19, found on B-cell lineages, and B-cell maturation antigen (BCMA), found on plasma cells and plasmablasts. Utilizing an OR gate logic, the CAR T cells are activated upon encountering either antigen, facilitating a more comprehensive depletion of pathogenic B-cell populations than single-target therapies. It is currently being investigated in Phase 1 clinical trials for conditions such as systemic lupus erythematosus, systemic sclerosis, and idiopathic inflammatory myopathy.
