Autologous CD19-specific CAR T-cells (also referred to as CAR19 T-cells) is an investigational cell-based gene therapy developed by the Blood Transplant and Cell Therapies Program at Westmead Hospital, in collaboration with the Westmead Institute for Medical Research and the University of Sydney. The therapy consists of the patient's own T-cells genetically engineered ex vivo using the non-viral piggyBac transposon system to express a chimeric antigen receptor (CAR) targeting CD19, a surface antigen highly expressed on B-cell malignancies. It is being evaluated in a Phase 1 clinical trial (The Auto-CAR19 Trial, ACTRN12619000079145) in Australia for the treatment of relapsed, refractory, or persistent CD19-positive B-cell malignancies, including acute lymphoblastic leukemia (ALL), chronic lymphocytic leukemia (CLL), and non-Hodgkin lymphoma (NHL).
