Overview

Autologous CD19-STAR-T cell is a type of adoptive cell therapy that utilizes a patient's own T cells, genetically engineered to express a Synthetic T cell receptor and Antigen Receptor (STAR) targeting the CD19 protein. Unlike conventional chimeric antigen receptors (CARs), the STAR receptor replaces the native T cell receptor (TCR) alpha/beta variable domains with antibody-derived variable regions, integrating directly into the endogenous TCR/CD3 complex. This design allows for signal transduction through native TCR molecules upon CD19 antigen engagement, leading to robust T cell activation, proliferation, and cytotoxic activity against CD19-expressing cells, independent of MHC presentation. This approach aims to improve T cell function and persistence compared to earlier CAR designs.
