Overview

Autologous CD34+ hematopoietic stem cells are progenitor cells harvested from a patient's own peripheral blood, typically following mobilization with agents like G-CSF or plerixafor. These cells are used in the context of autologous stem cell transplantation (ASCT) to reconstitute the bone marrow and immune system after a patient has received myeloablative high-dose chemotherapy, such as the BEAM regimen. In clinical research led by Memorial Sloan Kettering Cancer Center specifically for diffuse large B-cell lymphoma (DLBCL), investigators are studying the impact of the CD34+ cell dose on patient outcomes. The research evaluates whether a higher dose (6-8 x 10^6 cells/kg) compared to a standard dose (3-4 x 10^6 cells/kg) leads to faster recovery of the absolute lymphocyte count (ALC), which is associated with improved progression-free survival in relapsed or refractory DLBCL patients.
