Overview

Autologous CD4+ antigen-specific T-cells are an investigational adoptive cellular immunotherapy developed for the treatment of metastatic melanoma. This personalized therapy involves isolating a patient's own CD4+ T-lymphocytes through leukapheresis and expanding them ex vivo into clones that specifically recognize tumor-associated antigens (TAAs) such as MART-1, tyrosinase, or gp100. While CD8+ T cells are the traditional effectors in oncology, these specialized CD4+ "helper" T cells are designed to orchestrate a robust immune response by releasing cytokines, promoting the persistence of CD8+ cytotoxic T cells, and in some cases, exerting direct cytolytic activity against MHC class II-positive tumor cells. The treatment protocol typically involves dose-escalation to determine the maximum tolerated dose and persistence of the infused clones in vivo.
