Autologous CD40L and IL-2-expressing B-CLL cell vaccine is an experimental cellular immunotherapy developed by Baylor College of Medicine for the treatment of B-cell chronic lymphocytic leukemia (B-CLL). The vaccine is produced by modifying a patient's own leukemic B-cells to express two immune-modulating proteins: CD40 ligand (CD40L) and Interleukin-2 (IL-2). CD40L (CD154) is a costimulatory molecule that, when expressed on the tumor cells, activates antigen-presenting cells via the CD40 receptor, enhancing the presentation of tumor-associated antigens. IL-2 is a cytokine that promotes the activation and expansion of cytotoxic T-lymphocytes. By combining these two signals, the vaccine aims to overcome the characteristic immune tolerance of CLL and induce a robust, long-lasting systemic immune response against the cancer cells.
