Overview

Autologous CD5 CAR T-cells are a first-in-class chimeric antigen receptor T-cell therapy targeting CD5, a common surface marker expressed on normal and malignant T cells. This therapy involves collecting a patient's own T cells, genetically modifying them to express a CD5-directed CAR, and reinfusing them to target CD5-positive malignancies. The CD5 CAR produces minimal fratricide by downmodulating CD5 protein levels in transduced T cells while retaining strong cytotoxicity against CD5+ malignant cells. This approach allows for selective targeting of malignant cells while preserving some normal T-cell function, addressing a key challenge in treating T-cell malignancies where shared antigens between normal and malignant T cells can lead to fratricide or profound immunodeficiency.
