Overview

Autologous CMV-specific cytotoxic T-lymphocytes (CTLs) is an investigational adoptive cellular immunotherapy developed by the Baylor College of Medicine for the treatment of glioblastoma multiforme (GBM). The therapy involves harvesting a patient's own T cells and expanding them ex vivo to specifically recognize cytomegalovirus (CMV) antigens, such as pp65 and IE1, which are frequently expressed in GBM tumor cells but not in healthy brain tissue. This expansion is typically achieved using antigen-presenting cells (monocytes or dendritic cells) transduced with an adenoviral vector encoding these CMV proteins. Once infused back into the patient, these "trained" CTLs are designed to target and eliminate CMV-expressing tumor cells, potentially bypassing the immunosuppressive environment of the tumor and providing a targeted anti-tumor immune response.
