Overview

Autologous cultured epidermis (ACE) is a tissue-engineered regenerative medicine product composed of a patient's own keratinocytes expanded in vitro into cohesive epithelial sheets. Developed primarily for the treatment of extensive skin loss, the process involves isolating keratinocytes from a small skin biopsy and culturing them, typically using a 3T3 feeder layer, to create a multi-layered epidermal membrane. Once transplanted onto a wound bed, these sheets integrate with the host tissue to permanently restore the epidermal barrier. This therapy is particularly critical for patients with severe burns covering a large percentage of the total body surface area (TBSA), where donor sites for conventional skin grafts are insufficient. Beyond burns, it is also utilized for congenital conditions like giant melanocytic nevi and genetic disorders such as epidermolysis bullosa.
