Autologous cultured fibroblasts on amniotic membrane is an experimental cell therapy developed by the Royan Institute for the treatment of mitten hand deformity (syndactyly) in patients with Recessive Dystrophic Epidermolysis Bullosa (RDEB). The treatment involves isolating dermal fibroblasts from a patient's own skin biopsy, expanding them in vitro, and seeding them onto a human amniotic membrane, which serves as a biological scaffold. Following the surgical release of finger adhesions, this cell-seeded membrane is transplanted onto the denuded areas of the hand. The therapy aims to provide a biological dressing that facilitates healing and addresses the underlying deficiency of Type VII collagen, a protein essential for anchoring the epidermis to the dermis, which is defective in RDEB due to mutations in the COL7A1 gene.
