Autologous cultured revertant keratinocytes are an investigational cell therapy product designed for the treatment of Epidermolysis Bullosa (EB), including Junctional (JEB) and Dystrophic (DEB) forms. The therapy utilizes the biological phenomenon of revertant mosaicism, in which a patient's own skin cells naturally acquire a second-site mutation that corrects a germline pathogenic defect, creating patches of healthy skin. In this therapeutic approach, keratinocytes are harvested from these naturally corrected areas, expanded in vitro to form cohesive epidermal sheets, and then surgically grafted onto the patient’s chronic wounds or areas of high fragility. This method allows for the restoration of critical structural proteins in the dermo-epidermal junction, such as laminin-332 or type VII collagen, without the need for viral-mediated gene editing or exogenous genetic modification. The resulting grafts are autologous and thus immune-compatible, providing a durable solution for wound healing and skin integrity in severely affected patients.
