Autologous cytokine-induced killer (CIK) cells are an investigational cell-based immunotherapy being developed by Prof. Franca Fagioli at the Sant'Anna OIRM Hospital for the treatment of advanced, relapsed, and non-resectable sarcomas. CIK cells are ex vivo expanded T lymphocytes that exhibit a mixed T-NK phenotype (CD3+CD56+) and possess potent, MHC-unrestricted antitumor activity. The cytotoxic effect is primarily mediated through the NKG2D receptor, which recognizes stress-inducible ligands such as MIC A/B and ULBPs expressed on the surface of sarcoma cells, including chemoresistant cancer stem-like cells. This therapy is currently being evaluated in Phase I clinical trials to determine the maximum tolerated dose and safety profile in pediatric and adult patients with bone and soft-tissue sarcomas.
