Overview

Autologous Cytomegalovirus-specific Cytotoxic T-lymphocytes (CMV-CTLs) represent an adoptive cell therapy where a patient's own (autologous) T-lymphocytes are collected, expanded ex vivo, and specifically trained to recognize and eliminate cells infected with Cytomegalovirus (CMV). These T-cells, primarily CD4+ and CD8+ cytotoxic T-lymphocytes, target CMV antigens such as pp65 and IE1. The therapy aims to restore or enhance CMV-specific immunity, particularly in immunocompromised patients, such as those undergoing hematopoietic stem cell transplantation (HSCT) or solid organ transplant, who are at high risk for CMV reactivation or infection. It has also been investigated as a consolidative treatment for recurrent glioblastoma multiforme (GBM) due to the expression of CMV antigens in GBM tissues. The mechanism of action involves the direct lysis of CMV-infected cells, and it is being explored as a pre-emptive therapy or for the treatment of refractory CMV infections.
