Autologous dendritic cell vaccines are a form of personalized cancer immunotherapy designed to stimulate a patient's own immune system to recognize and destroy tumor cells. The manufacturing process involves harvesting a patient's monocytes via leukapheresis, which are then differentiated ex vivo into mature dendritic cells (DCs). These DCs are subsequently loaded with tumor-associated antigens (TAAs) derived from various sources, such as autologous tumor cell lines, irradiated tumor cells, or mRNA-encoded neoantigens. Once re-infused into the patient, these professional antigen-presenting cells migrate to lymphoid organs where they prime and activate T-cell mediated anti-tumor immunity. This platform approach aims to overcome tumor-induced immunosuppression by presenting a broad repertoire of patient-specific antigens, including neoantigens, in a highly immunogenic context.
