This autologous dendritic cell-based cancer vaccine is a personalized biologic immunotherapy developed by Duke University for the treatment of metastatic prostate cancer. The vaccine is produced by isolating a patient's own dendritic cells and loading them with tumor-associated antigens, either through transfection with mRNA encoding prostate-specific antigen (PSA) or by pulsing the cells with autologous tumor-derived RNA. Once administered, these mature dendritic cells present the antigens to the patient's immune system, specifically aiming to stimulate PSA-specific cytotoxic T lymphocyte (CTL) responses to target and eliminate metastatic prostate tumor cells. Phase I clinical trials have demonstrated the vaccine's safety and its ability to induce specific T-cell responses and provide preliminary clinical benefits, such as reduced PSA velocity.
