The autologous dendritic cell vaccine loaded with allogeneic DIPG tumor lysate is an investigational cellular immunotherapy developed by Fundació Sant Joan de Déu for the treatment of newly diagnosed diffuse intrinsic pontine glioma (DIPG). The vaccine is produced by harvesting a patient's monocytes via leukapheresis, maturing them into dendritic cells ex vivo, and pulsing them with a lysate derived from a pool of allogeneic, K27M-mutated DIPG tumor cell lines. This process allows the dendritic cells to present tumor-specific antigens to the patient's immune system. Administered intradermally, the vaccine is designed to induce a robust T-cell mediated immune response against the aggressive brainstem tumor. Clinical evaluation in a Phase Ib study (NCT02840123) demonstrated safety and the induction of specific immune responses in both peripheral blood and cerebrospinal fluid.
