Autologous dendritic cells electroporated with mRNA encoding gp100 and tyrosinase is an investigational **autologous dendritic-cell cancer vaccine** for melanoma. Patient-derived monocyte-derived dendritic cells are loaded ex vivo with messenger RNA encoding the melanoma-associated differentiation antigens premelanosome protein gp100 and tyrosinase, enabling endogenous antigen expression and presentation through major histocompatibility complex pathways. After administration, the antigen-presenting dendritic cells are intended to prime and expand gp100-specific and tyrosinase-specific CD4-positive and CD8-positive T-cell responses against melanoma cells expressing these antigens. This approach was evaluated by Radboud University Medical Center investigators in stage III and stage IV melanoma, including intranodal vaccination studies; keyhole limpet hemocyanin was used in some protocols as an immune-control antigen.
