Autologous dendritic cells encoding WT1, PRAME, and CMVpp65 is an investigational cell therapy designed as a cancer vaccine. It involves extracting dendritic cells from a patient (autologous), genetically modifying them to express or present specific tumor-associated antigens: Wilms' Tumor 1 (WT1), Preferentially expressed antigen in melanoma (PRAME), and Cytomegalovirus pp65 (CMVpp65). WT1 and PRAME are known leukemia-associated antigens (LAAs) and are overexpressed in various cancers, particularly acute myeloid leukemia (AML). CMVpp65 is a protein from the human cytomegalovirus (HCMV) that has been utilized as a tumor-specific antigen, especially in glioblastoma, where HCMV genes are implicated in tumor proliferation. The modified dendritic cells are then re-administered to the patient, where they function as antigen-presenting cells. Their primary mechanism of action is to stimulate the patient's immune system, specifically T-cell responses, to recognize and target cancer cells that express these antigens, thereby inducing an anti-tumor immune response.
