Autologous dendritic cells expressing TAT mRNA is an investigational therapeutic HIV-1 vaccine consisting of autologous monocyte-derived dendritic cells electroporated with mRNA encoding the early HIV-1 regulatory and accessory proteins Tat, Rev, and Nef. Developed through a collaboration between Erasmus MC and Vrije Universiteit Brussel, the vaccine is designed to present these viral antigens to the patient's immune system to elicit or enhance antigen-specific CD4+ and CD8+ T-cell responses. By targeting proteins expressed early in the viral life cycle, the vaccine aims to improve immune control of HIV-1 replication, particularly in patients undergoing analytical treatment interruption (ATI) from combination antiretroviral therapy (cART). Clinical studies, such as the Phase I/IIa DCTRN trial, have demonstrated the safety and immunogenicity of this approach.
