Overview

The autologous dendritic cells HepG2-pulsed vaccine is an investigational cell-based immunotherapy developed by the University of Birmingham for the treatment of hepatocellular carcinoma (HCC). The vaccine consists of mature autologous dendritic cells derived from the patient's peripheral blood, which are pulsed ex vivo with a total cell lysate prepared from the HepG2 human hepatocellular carcinoma cell line. This process loads the dendritic cells with a broad repertoire of tumor-associated antigens (TAAs) expressed by HCC cells. Upon intravenous administration, these primed dendritic cells are intended to stimulate a robust, antigen-specific cytotoxic T lymphocyte (CTL) response against the patient's tumor. In clinical studies, the vaccine has been evaluated in combination with preconditioning cyclophosphamide and transarterial chemoembolisation (TACE), demonstrating improved progression-free survival and a manageable safety profile.
