Overview

Autologous dendritic cells infected with fowlpox-CEA-TRICOM is an ex vivo-generated cellular immunotherapy designed to treat cancers expressing carcinoembryonic antigen (CEA). The therapy involves harvesting a patient's own dendritic cells (DCs) and infecting them with a recombinant fowlpox virus (rF) vector. This vector encodes the CEA antigen along with a triad of T-cell costimulatory molecules (TRICOM): B7-1 (CD80), ICAM-1 (CD54), and LFA-3 (CD58). Once re-infused into the patient, these engineered DCs present CEA peptides to the immune system. The TRICOM components act synergistically to enhance the activation, signaling, and expansion of CEA-specific cytotoxic T lymphocytes (CTLs), overcoming the immune tolerance often associated with tumor-associated antigens. This approach was primarily developed by researchers at the National Cancer Institute (NCI) for indications such as metastatic colorectal and prostate cancers.
