Autologous dendritic cells loaded with allogeneic brain tumor stem cells is an investigational cellular immunotherapy designed to treat glioblastoma multiforme. This therapeutic vaccine consists of a patient's own (autologous) dendritic cells that are harvested via leukapheresis, matured ex vivo, and pulsed (loaded) with antigens derived from a standardized, allogeneic (third-party) brain tumor stem cell (BTSC) line. BTSCs are a subpopulation of tumor cells characterized by self-renewal and resistance to standard radiochemotherapy; they are believed to be the primary drivers of tumor recurrence. By loading dendritic cells with the comprehensive antigenic profile of these allogeneic stem cells, the vaccine aims to prime and activate the patient's T-lymphocytes to recognize and eliminate the patient's own endogenous brain tumor stem cells. The use of an allogeneic cell line ensures a consistent and characterized source of cancer stem cell-associated antigens without the need for patient-specific tumor tissue cultivation.
