Autologous dendritic cells loaded with allogeneic mesothelioma tumor cell lysate (commercially known as MesoPher) is an investigational cell-based immunotherapy developed by Erasmus Medical Center and its spin-off company, Amphera BV. The therapy is prepared by collecting patient monocytes via leukapheresis, differentiating them ex vivo into dendritic cells (DCs), and loading them with PheraLys—a proprietary, clinical-grade allogeneic lysate derived from mesothelioma cell lines containing a broad repertoire of tumor-associated antigens (TAAs). MesoPher is designed to stimulate a robust, tumor-specific CD4+ and CD8+ T-cell response to prevent tumor recurrence and combat tumor-immune escape. It has been evaluated in clinical trials for the treatment of malignant pleural mesothelioma (including the Phase II/III DENIM trial as maintenance therapy) and peritoneal mesothelioma, as well as pancreatic ductal adenocarcinoma (including the Phase II REACTIVE trial and Phase I REACTIVE-2 trial in combination with the CD40 agonist mitazalimab).
