Autologous dendritic cells loaded with autologous tumor RNA is a personalized immunotherapy approach that uses a patient's own dendritic cells (DCs) loaded with RNA extracted from their tumor cells to create a therapeutic cancer vaccine. This treatment works by enhancing the immune system's ability to recognize and attack cancer cells. The process involves: 1. Collecting tumor cells from the patient 2. Extracting RNA from these tumor cells 3. Isolating monocytes from the patient's peripheral blood 4. Culturing these monocytes with specific cytokines (GM-CSF and IL-4) to generate dendritic cells 5. Loading the dendritic cells with the tumor RNA through electroporation or co-incubation 6. Administering the loaded dendritic cells back to the patient as a vaccine The loaded dendritic cells present tumor antigens to T cells, stimulating a tumor-specific immune response. This approach has been investigated in various cancers including melanoma, glioblastoma, prostate cancer, and renal cell carcinoma.
