Autologous dendritic cells loaded with autologous tumor RNA is a personalized, patient-specific cancer vaccine classified as an Advanced Therapy Medicinal Product (ATMP). Developed by the Experimentelle Immuntherapie unit at Universitätsklinikum Erlangen, the vaccine consists of autologous, mature, monocyte-derived dendritic cells that are transfected with RNA extracted and amplified from the patient's own resected tumor tissue. The primary indication is high-risk, monosomy 3 uveal melanoma. The mechanism involves the presentation of tumor-specific antigens by the dendritic cells to the patient's immune system, thereby inducing a cytotoxic T lymphocyte (CTL) response against the tumor. A Phase III trial (NCT01983748) evaluating this adjuvant therapy versus observation was completed in 2024.
