Autologous dendritic cells pulsed with allogeneic tumour cell lysate is an investigational cellular immunotherapy developed by Instituto Oncologico Dr. Rosell for the treatment of small cell lung cancer (SCLC). This personalized vaccine is produced by isolating monocytes from a patient's peripheral blood, differentiating them into mature dendritic cells ex vivo, and then 'pulsing' them with a lysate derived from a standardized pool of allogeneic SCLC cell lines (such as H69, H128, and H209). The resulting dendritic cells, which present a broad array of tumor-associated antigens, are administered intradermally to the patient to stimulate a systemic, T-cell-mediated anti-tumor immune response. This approach aims to overcome the difficulty of obtaining sufficient autologous tumor tissue for vaccine preparation in SCLC patients while providing a diverse set of antigens to minimize immune escape.
