Autologous dendritic cells pulsed with tumor lysate antigen vaccine is a personalized immunotherapy in which a patient's own (autologous) dendritic cells are harvested and then exposed ex vivo to antigens derived from the patient's own tumor lysate. The resulting "pulsed" or "loaded" dendritic cells are then re-administered to the patient as a therapeutic cancer vaccine. The mechanism of action involves the presentation of a broad array of patient-specific tumor antigens by mature dendritic cells to T lymphocytes, thereby priming and activating an anti-tumor immune response. This approach aims to overcome the heterogeneity of tumors by targeting multiple antigens present in each individual’s cancer[4][5]. It has been most extensively studied in glioblastoma (notably as DCVax-L), where it has shown statistically significant improvements in overall survival when added to standard-of-care therapy[2][4][5]. Other indications under investigation include soft tissue sarcoma and other solid tumors[6][7].
