Autologous dendritic cells transduced with Ad encoding NS3 is a personalized cell-based immunotherapy developed as a therapeutic vaccine for chronic hepatitis C virus (HCV) infection. The therapy involves isolating monocyte-derived dendritic cells (MoDC) from the patient, then genetically modifying these cells ex vivo using a recombinant adenoviral vector that encodes the nonstructural protein 3 (NS3) of HCV. The modified DCs are matured and re-infused into the patient to stimulate an immune response specifically targeting HCV-infected hepatocytes. The mechanism of action centers on presenting HCV NS3 antigens to T-cells, thereby inducing both CD4+ and CD8+ T-cell responses against the virus. Clinical studies have shown that this approach is safe and can induce antigen-specific immune responses, though antiviral efficacy has been limited in early-phase trials[1][2][5]. This strategy was primarily investigated before direct-acting antivirals became standard care for HCV.
