Autologous dendritic cells transfected with leukemia-associated antigen RNA is an investigational cell-based immunotherapy developed by the Hospital of Ludwig Maximilians University Munich. The therapy involves generating autologous monocyte-derived dendritic cells (DCs) ex vivo from the patient's own CD14+ monocytes, which are then transfected via electroporation with in vitro-transcribed mRNA encoding leukemia-associated antigens (LAAs), specifically Wilms' Tumor protein 1 (WT1) and Preferentially Expressed Antigen in Melanoma (PRAME). These antigen-loaded mature DCs are re-administered to the patient to stimulate an AML-specific T-cell immune response. It was evaluated in a Phase 1 clinical trial (NCT01734304) for the treatment of acute myeloid leukemia (AML) in adult patients not eligible for allogeneic stem cell transplantation.
