Overview

Autologous Epstein-Barr virus-specific cytotoxic T lymphocytes (EBV-CTLs) represent an adoptive cell therapy where a patient's own T cells are expanded and directed to specifically target and eliminate cells infected with the Epstein-Barr virus (EBV). This therapeutic approach has demonstrated safety and efficacy in the treatment of EBV-associated posttransplantation lymphoproliferative disorders (PTLDs) in solid organ transplant recipients. Research is ongoing to enhance the therapy, including modifications to make these cells resistant to immunosuppressive drugs such as tacrolimus (FK506) by knocking down the expression of FKBP12. This modification aims to improve the long-term persistence of transferred EBV-CTLs and enhance EBV-specific immune surveillance in patients at high risk of PTLD after transplantation.
