Overview

This investigational adoptive cellular immunotherapy consists of autologous natural killer (NK) cells that are collected from a patient, expanded, and activated ex vivo using a specialized feeder cell system expressing membrane-bound interleukin-21 (IL-21) and 4-1BB ligand. Developed by the New Approaches to Neuroblastoma Therapy (NANT) Consortium, this therapy is specifically designed for the treatment of relapsed or refractory neuroblastoma. The expansion process is intended to significantly increase the number and cytotoxic potential of the NK cells. In clinical trials, these cells are administered intravenously in combination with the anti-GD2 monoclonal antibody dinutuximab (ch14.18), with or without the immunomodulatory agent lenalidomide. The therapeutic mechanism involves direct NK cell-mediated lysis of tumor cells and the enhancement of antibody-dependent cellular cytotoxicity (ADCC) against GD2-expressing neuroblastoma cells.
