Autologous formalin-fixed tumor vaccine (AFTV) is a personalized cancer immunotherapy consisting of formalin-fixed tumor tissue fragments or cells derived from a patient's own resected malignancy. The formalin fixation process inactivates the tumor cells while preserving the structural integrity of a broad array of unidentified tumor-associated antigens (TAAs). When administered intradermally, typically alongside adjuvants such as granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-2 (IL-2), the vaccine stimulates the patient's immune system to recognize and attack residual tumor cells. Clinical studies have primarily focused on preventing recurrence in glioblastoma multiforme (GBM) and hepatocellular carcinoma (HCC), where it has demonstrated the ability to induce delayed-type hypersensitivity (DTH) responses and glypican-3-specific cytotoxic T lymphocyte activity.
