Autologous heat shock protein 70-peptide complexes (Hsp70PC) is a personalized cancer vaccine developed by UConn Health for the treatment of chronic myelogenous leukemia (CML). The vaccine is produced by purifying heat shock protein 70 (HSP70) complexed with intracellular peptides from the patient's own leukopheresed peripheral blood mononuclear cells. HSP70 serves as a molecular chaperone that carries a broad repertoire of tumor-specific antigenic peptides. When administered intradermally, these complexes are captured by antigen-presenting cells, such as dendritic cells, which then cross-present the CML-specific antigens to T cells. Additionally, the vaccine activates natural killer (NK) cells, collectively mounting a targeted immune response against the CML cells. In clinical studies, it has been evaluated in combination with tyrosine kinase inhibitors like imatinib mesylate.
