Overview

This combination therapy consists of autologous hematopoietic stem cell transplantation (Auto-HSCT) followed by the infusion of inaticabtagene autoleucel (CNCT19), a CD19-directed chimeric antigen receptor (CAR) T-cell therapy. Inaticabtagene autoleucel is an autologous CAR-T product utilizing a single-chain variable fragment (scFv) derived from the HI19α clone, with a 4-1BB costimulatory domain and a CD3-zeta signaling domain. The regimen is being evaluated for adult patients with high-risk B-cell acute lymphoblastic leukemia (B-ALL) who are in complete remission, aiming to eliminate minimal residual disease (MRD) and reduce the risk of relapse. The CAR-T cells are typically administered approximately 7 days after the stem cell transplant to provide a targeted anti-leukemic effect during the post-transplant recovery phase.
