Overview

Autologous hematopoietic stem cells (HSCs) are utilized as a supportive cellular therapy by the University of Florida in the ADAGiO clinical trial (NCT05704504) for patients with recurrent or progressive WHO grade 2 and 3 oligodendroglioma. In this therapeutic context, CD34+ HSCs are harvested from the patient and administered as a single intravenous infusion following lymphodepleting conditioning. The primary role of these cells is to provide hematopoietic rescue and facilitate the engraftment and homeostatic expansion of ex vivo expanded tumor-reactive T cells (TTRNA-xALT). This approach is part of a complex immunotherapy regimen that also includes total tumor RNA (TTRNA)-loaded dendritic cell vaccines and IDH1/2 inhibitors, aimed at overcoming the immunosuppressive microenvironment of gliomas.
