Overview

Autologous hematopoietic stem cells (HSCs) are patient-derived multipotent progenitor cells used to restore bone marrow function following high-dose, myeloablative chemotherapy. In the context of clinical research at M.D. Anderson Cancer Center, these cells are utilized for patients with multiple myeloma or primary amyloidosis undergoing autologous stem cell transplantation (ASCT). The process involves mobilizing HSCs from the patient's peripheral blood using agents like granulocyte-colony stimulating factor (G-CSF), collecting them via apheresis, and re-infusing them after the administration of conditioning agents such as melphalan. A specific Phase II trial (NCT01142336) investigated whether higher doses of infused CD34+ cells (10-15 x 10^6 cells/kg) could reduce transplant-induced symptom burden, including fatigue and gastrointestinal distress, and improve quality of life compared to standard doses (4-6 x 10^6 cells/kg).
