Autologous HER2-targeted CAR-T cell therapy is an investigational cell-based immunotherapy developed by the National University Hospital, Singapore. The treatment involves the ex vivo genetic modification of a patient's own T-lymphocytes to express a chimeric antigen receptor (CAR) that specifically recognizes the Human Epidermal Growth Factor Receptor 2 (HER2/neu), a protein overexpressed in various malignancies. Once re-infused into the patient following a lymphodepleting chemotherapy regimen (typically fludarabine and cyclophosphamide), these engineered T-cells bind to HER2-positive tumor cells, leading to T-cell activation and targeted cell lysis. It is currently being evaluated in Phase Ib/II clinical trials, often in combination with the monoclonal antibody trastuzumab, for the treatment of HER2-positive advanced solid tumors, including breast and gastric cancers.
