Autologous IFNγ-stimulated mesenchymal stromal cells, also known as MSC(M), are an investigational cell therapy being developed for the treatment of radiation-induced xerostomia (RIX) in head and neck cancer patients. The manufacturing process involves harvesting bone marrow-derived mesenchymal stromal cells (MSCs) from the patient, expanding them in vitro, and "licensing" them with interferon-gamma (IFNγ). This stimulation enhances the cells' immunomodulatory and regenerative properties, notably increasing the expression of indoleamine 2,3-dioxygenase (IDO). The cryopreserved cells are thawed and injected directly into the patient's submandibular glands to promote tissue repair and restore salivary function. The therapy is currently being evaluated in the Phase 1 MARSH trial (NCT04489732) at the University of Wisconsin-Madison.
