Overview

Autologous interleukin-2-secreting neuroblastoma vaccine is an investigational personalized cellular immunotherapy designed for the treatment of high-risk or recurrent neuroblastoma. The therapeutic process involves harvesting neuroblastoma cells from a patient, which are subsequently genetically modified in vitro—often using a retroviral or adenoviral vector—to express and secrete the cytokine interleukin-2 (IL-2). After being irradiated to prevent further proliferation, these modified cells are administered back to the patient. The mechanism of action relies on the localized secretion of IL-2 at the site of administration or within the tumor microenvironment, which serves to recruit, activate, and expand tumor-specific cytotoxic T-lymphocytes and natural killer (NK) cells. This approach aims to bypass tumor-induced immune suppression and generate a systemic, durable anti-tumor immune response against neuroblastoma-specific antigens.
