Overview

Autologous invariant natural killer T (iNKT) cells are an adoptive cell therapy being developed by Beijing YouAn Hospital for the treatment of hepatocellular carcinoma (HCC). The therapeutic process involves isolating iNKT cells from a patient's peripheral blood mononuclear cells, expanding them ex vivo, and pulsing them with the glycolipid alpha-galactosylceramide (α-GalCer). These cells are a unique population of CD1d-restricted T cells that bridge the innate and adaptive immune systems. Upon reinfusion, they exert potent antitumor activity by rapidly producing Type 1 helper (Th1) cytokines, such as interferon-gamma, and releasing cytotoxic granules containing perforin and granzyme B to directly target malignant cells. The therapy is currently being evaluated in Phase 2 clinical trials, often in combination with multi-kinase inhibitors like regorafenib and immune checkpoint inhibitors, for patients with progressed HCC.
