Overview

Autologous lymphocyte infusion (ALI) is an adoptive cell therapy developed by the H. Lee Moffitt Cancer Center and Research Institute for the treatment of metastatic melanoma. The procedure involves the ex vivo expansion of a patient's own lymphocytes, which are then re-infused into the patient to bolster the immune system's ability to recognize and destroy tumor cells. In clinical protocols (such as NCT00338377), ALI is typically administered following a lymphodepleting conditioning regimen of fludarabine and in combination with a dendritic cell vaccine pulsed with melanoma-associated antigens, including MART-1, gp100, tyrosinase, and NY-ESO-1. This multi-component approach is designed to create a favorable environment for the infused lymphocytes to proliferate and mount a targeted T-cell response against malignant melanoma cells.
