Overview

**Autologous lymphokine-activated killer cells** are a patient-specific adoptive cellular immunotherapy developed by Hoag Memorial Hospital Presbyterian for glioblastoma multiforme. Peripheral-blood lymphocytes or peripheral-blood mononuclear cells are collected from the patient and activated ex vivo with interleukin-2 for several days to generate lymphokine-activated killer cells, then placed directly into the surgically exposed tumor cavity. The product is intended to provide local, non-antigen-restricted immune-mediated cytotoxicity against residual glioblastoma cells. Hoag evaluated intralesional autologous LAK cells in Phase II studies in primary and recurrent glioblastoma; the published primary-glioblastoma study reported that treatment was well tolerated and supported further randomized Phase II evaluation.
