Overview

A patient-specific cancer DNA vaccine designed for the treatment of B-cell lymphomas such as lymphoplasmacytic lymphoma. The vaccine is created by isolating the variable regions (VH and VL) of the immunoglobulin from a patient's own tumor cells, constructing a single-chain variable fragment (scFv), and fusing it to the human chemokine CCL20 (macrophage inflammatory protein 3 alpha, MIP-3α). This fusion gene is then inserted into a plasmid vector to create a personalized DNA vaccine. Upon intradermal administration, the encoded fusion protein targets antigen-presenting cells expressing CCR6 via CCL20-mediated chemotaxis and presents tumor-specific idiotype antigens to stimulate an anti-tumor immune response[1][2][7]. The approach aims to enhance dendritic cell uptake and presentation of tumor antigens, thereby inducing both humoral and cellular immunity against residual or relapsing lymphoma.
